VirCap ® InertShell is a composite chromatography medium with a core-shell structure that combines volume exclusion, anion exchange, and hydrophobic interactions. After multiple tests, it has demonstrated excellent performance in the purification of inactivated viruses.
VirCap ® The matrix of InertShell is polyacrylate microspheres, and its microsphere structure and separation principle are shown in Figure 1. The shell of the microspheres is a neutral hydrophilic layer, which has non-specific adsorption for proteins; At the same time, the shell pore size (50-100 nm) is smaller than the core pore size (200-500 nm), and its shell thickness is about 0.5-1.0 μ m. The shell can prevent proteins with a molecular weight greater than 700 kDa from entering the core layer. Therefore, during the chromatography process, large-sized inactivated viruses cannot enter the interior of the microspheres, so they quickly flow through and are collected. The core part of the medium is activated by the octylamine group, thus achieving dual functions of anion exchange and hydrophobic interaction, which can quickly capture protein molecules with a molecular weight less than 700 kDa, and effectively remove impurities such as host cell proteins, DNA fragments, endotoxins, and albumin in the purification system.
